چکیده

Conclusions Use of the drug tunicamycin has greatly facilitated study of many different aspects of immunoglobulin biosynthesis. The most striking conclusion is that blocking glycosylation does not prevent synthesis of immunoglobulin heavy chains either directly or by feedback; non-glycosylated heavy chains are able to assemble into immunoglobulin molecules capable of being secreted. Since glycosylation affects overall protein threedimensional structure, differences in the structure of nonglycosylated immunoglobulins might be responsible for the failure of non-glycosylated IgM to be transferred to the cell surface, alterations in the details of assembly and secretion of the non-glycosylated immunoglobulins. Such changes in structure in the non-glycosylated molecules might also affect function, and studies on the function of non-glycosylated antibodies should prove rewarding.

Effect of N-glycosylation inhibition on the synthesis and processing of classical swine fever virus glycoproteins.

Classical swine fever virus (CSFV) is often used as a surrogate model in molecular studies of the closely related hepatitis C virus. In this report we have examined the effect of the inhibition of glycosylation on the survival and maturation of CSFV. Viral glycoproteins (E(rns), E1, E2) form biologically active complexes - homo- and heterodimers, which are indispensable for viral life cycle. Th...

The Relationship of Secretion and Activity of Recombinant Factor IX with N-Glycosylation

Background:  Human coagulation factor IX (hFIX) is a glycoprotein with two N-glycosylation sites at the activation peptide. Since the activation peptide is removed in mature hFIX, the exact role of N-glycosylation is unclear. To investigate the role of N-glycosylation in the secretion and activity of hFIX, we inhibited N-glycosylation by tunicamycin in the stable Human Embryonic Kidney (HEK)- c...

Authentic processing and targeting of active maize auxin-binding protein in the baculovirus expression system.

The major auxin-binding protein (ABP1) from maize (Zea mays L.) has been expressed in insect cells using the baculovirus expression system. The recombinant protein can be readily detected in total insect cell lysates by Coomassie blue staining on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Our data suggest that ABP1 is processed similarly in both insect cells and maize...

Glycosylation is essential for biosynthesis of functional gastric H+,K+-ATPase in insect cells.

The role of N-linked glycosylation in the functional properties of gastric H+,K+-ATPase has been examined with tunicamycin and I-deoxymannojirimycin, inhibitors in glycoprotein biosynthesis and glycoprotein processing respectively. Tunicamycin completely abolished both K+-stimulated and 3-(cyanomethyl)-2-methyl-8-(phenylmethoxy)-imidazo[1,2a]pyridine (SCH 28080)-sensitive ATPase activity and SC...

Effect of tunicamycin on the biogenesis of hepatitis C virus glycoproteins.

Hepatitis C virus (HCV) infects humans, with a prevalence around 3% of population, causing acute and chronic hepatitis and hepatocellular carcinoma. We studied the effect of inhibition of glycosylation on the assembly of the HCV particle. HCV possesses two envelope glycoproteins E1 and E2 that are highly modified by N-glycans. These glycan residues are crucial for viral entry and maturation of ...
